RESUMO

Acute mesenteric ischemia (AMI) is a clinically significant vascular and gastrointestinal condition, which is closely related to the blood supply of the small intestine. Unfortunately, it is still challenging to properly discriminate small intestinal tissues with different degrees of ischemia. In this study, hyperspectral imaging (HSI) was used to construct pseudo-color images of oxygen saturation about small intestinal tissues and to discriminate different degrees of ischemia. First, several small intestine tissue models of New Zealand white rabbits were prepared and collected their hyperspectral data. Then, a set of isosbestic points were used to linearly transform the measurement data twice to match the reference spectra of oxyhemoglobin and deoxyhemoglobin, respectively. The oxygen saturation was measured at the characteristic peak band of oxyhemoglobin (560 nm). Ultimately, using the oxygenated hemoglobin reflectance spectrum as the benchmark, we obtained the relative amount of median oxygen saturation in normal tissues was 70.0 %, the IQR was 10.1 %, the relative amount of median oxygen saturation in ischemic tissues was 49.6 %, and the IQR was 14.6 %. The results demonstrate that HSI combined with the oxygen saturation computation method can efficiently differentiate between normal and ischemic regions of the small intestinal tissues. This technique provides a powerful support for internist to discriminate small bowel tissues with different degrees of ischemia, and also provides a new way of thinking for the diagnosis of AMI.

RESUMO

The present research investigates the effectiveness of nano-emulsified coatings (C-1, C-2, and C-3) in preserving the kiwifruit at a temperature of 10 ± 2 °C with 90-95 % relative humidity (RH) for 30 days. The nano-emulsions were prepared from varied carboxymethyl cellulose (CMC) concentrations with different combinations of essential oils such as thyme, clove, and cardamom. Dynamic light scattering investigation with Zeta Sizer revealed that C-1, C-2, and C-3 nano-emulsions have nano sizes of 81.3 ± 2.3, 115.3 ± 4.2, and 63.2 ± 3.2 nm, respectively. The scanning electron microscopy images showed that the nanoemulsion of C-1 had homogenous spherical globules, C-2 had voids, and C-3 showed a non-porous structure with uniform dispersion. The X-ray diffraction analysis indicated that C-1, C-2, and C-3 nano-emulsion exhibited distinct crystallinity and peaks. The nano-emulsion C-1 had reduced crystallinity, while C-2 had lower intensity peaks, and C-3 had increased crystallinity. The results documented that compared to control kiwifruit samples, the samples coated with C-3 nano-emulsion have decreased weight loss, decay incidence, soluble solids, maturity index activity, ethylene production, total bacterial count, and increased titratable acid, and firmness attributes. The results of current research are promising and would be applicable in utilization in industrial applications.

RESUMO

A method for spectral reflectance factor reconstruction based on wideband multi-illuminant imaging was proposed, using a programmable LED lighting system and modified Bare Bones Particle Swarm Optimization algorithms. From a set of 16 LEDs with different spectral power distributions, nine light sources with correlated color temperatures in the range of 1924 K - 15746 K, most of them daylight simulators, were generated. Samples from three color charts (X-Rite ColorChecker Digital SG, SCOCIE ScoColor paint chart, and SCOCIE ScoColor textile chart), were captured by a color industrial camera under the nine light sources, and used in sequence as training and/or testing colors. The spectral reconstruction models achieved under multi-illuminant imaging were trained and tested using the canonical Bare Bones Particle Swarm Optimization and its proposed modifications, along with six additional and commonly used algorithms. The impacts of different illuminants, illuminant combinations, algorithms, and training colors on reconstruction accuracy were studied comprehensively. The results indicated that training colors covering larger regions of color space give more accurate reconstructions of spectral reflectance factors, and combinations of two illuminants with a large difference of correlated color temperature achieve more than twice the accuracy of that under a single illuminant. Specifically, the average reconstruction error by the method proposed in this paper for patches from two color charts under A + D90 light sources was 0.94 and 1.08 CIEDE2000 color difference units. The results of the experiment also confirmed that some reconstruction algorithms are unsuitable for predicting spectral reflectance factors from multi-illuminant images due to the complexity of optimization problems and insufficient accuracy. The proposed reconstruction method has many advantages, such as being simple in operation, with no requirement of prior knowledge, and easy to implement in non-contact color measurement and color reproduction devices.

RESUMO

PURPOSE: This study aimed to compare the efficacy and safety of ultrasound-guided RFA and MWA in the treatment of unifocal PTMC. METHODS: This retrospective study included 512 patients with 512 unifocal papillary thyroid microcarcinomas (PTMCs) who underwent RFA (n = 346) and MWA (n = 166) between January 2021 and December 2021. The volumes of the ablation areas were measured during follow-up, and the volume reduction rates were evaluated. The ablation duration, volume of hydrodissection, and ablation-related complications were also compared between the groups. RESULTS: All lesions received complete ablation and no local or distant recurrences were observed in the two groups. A larger volume of isolation liquid was used for RFA than for MWA (p = 0.000). Hoarseness occurred in seven patients who underwent RFA (p = 0.102). At the 1-week follow-up, the mean volume of the areas ablated by RFA was smaller than that of the areas ablated by MWA (p = 0.049). During follow-ups at months 3, 9, 12, 15, and 18, the mean volumes of the ablated areas were larger in the RFA group than in the MWA group (all, p < 0.05). The mean volume of the ablated lesions increased slightly at the 1-week follow-up and then decreased at 1 month after ablation in both groups. The absorption curve of the ablated lesions in the RFA group was similar to that in the MWA group. CONCLUSIONS: RFA and MWA are both efficient and safe methods for treating unifocal PTMC. They may be alternative techniques for patients who are not eligible or are unwilling to undergo surgery.

RESUMO

Acquiring large amounts of hyperspectral data of small intestinal tissue with real labels in the clinic is difficult, and the data shows inter-patient variability. Building an automatic identification model using a small dataset presents a crucial challenge in obtaining a strong generalization of the model. This study aimed to explore the performance of hyperspectral imaging and transfer learning techniques in the automatic identification of normal and ischemic necrotic sites in small intestinal tissue. Hyperspectral data of small intestinal tissues were collected from eight white rabbit samples. The transfer component analysis (TCA) method was performed to transfer learning on hyperspectral data between different samples and the variability of data distribution between samples was reduced. The results showed that the TCA transfer learning method improved the accuracy of the classification model with less training data. This study provided a reliable method for single-sample modelling to detect necrotic sites in small intestinal tissue .

RESUMO

Uncontrolled hemorrhage caused by trauma can easily lead to death. Efficient and safe hemostatic materials are an urgent and increasing need for hemostatic research. Following a trauma, wound healing is induced by various cellular mechanisms and proteins. Hemostatic biomaterials that can not only halt bleeding quickly but also provide an environment to promote wound healing have been the focus of research in recent years. Mussel-inspired nanoparticle composite hydrogels have been propelling the development of hemostatic materials owing to their unique advantages in adhesion, hemostasis, and bacteriostasis. This review summarizes the hemostatic and antimicrobial fundamentals of polydopamine (PDA)-based nanomaterials and emphasizes current developments in hemorrhage-related PDA nanomaterials. Moreover, it briefly discusses safety concerns and clinical application problems with PDA hemostatic nanomaterials.

RESUMO

Objective and automatic clinical discrimination of normal and necrotic sites of small intestinal tissue remains challenging. In this study, hyperspectral imaging (HSI) and unsupervised classification techniques were used to distinguish normal and necrotic sites of small intestinal tissues. Small intestinal tissue hyperspectral images of eight Japanese large-eared white rabbits were acquired using a visible near-infrared hyperspectral camera, and K-means and density peaks (DP) clustering algorithms were used to differentiate between normal and necrotic tissue. The three cases in this study showed that the average clustering purity of the DP clustering algorithm reached 92.07% when the two band combinations of 500-622 and 700-858 nm were selected. The results of this study suggest that HSI and DP clustering can assist physicians in distinguishing between normal and necrotic sites in the small intestine in vivo.

RESUMO

Background: Axillary lymph node dissection (ALND) can be safely avoided in women with T1 or T2 primary invasive breast cancer (BC) and one to two metastatic sentinel lymph nodes (SLNs). However, cancellation of ALND based solely on SLN biopsy (SLNB) may lead to adverse outcomes. Therefore, preoperative assessment of LN tumor burden becomes a new focus for ALN status. Objective: This study aimed to develop and validate a nomogram incorporating the radiomics score (rad-score) based on automated breast ultrasound system (ABUS) and other clinicopathological features for evaluating the ALN status in patients with early-stage BC preoperatively. Methods: Totally 354 and 163 patients constituted the training and validation cohorts. They were divided into ALN low burden (<3 metastatic LNs) and high burden (≥3 metastatic LNs) based on the histopathological diagnosis. The radiomics features of the segmented breast tumor in ABUS images were extracted and selected to generate the rad-score of each patient. These rad-scores, along with the ALN burden predictors identified from the clinicopathologic characteristics, were included in the multivariate analysis to establish a nomogram. It was further evaluated in the training and validation cohorts. Results: High ALN burdens accounted for 11.2% and 10.8% in the training and validation cohorts. The rad-score for each patient was developed based on 7 radiomics features extracted from the ABUS images. The radiomics nomogram was built with the rad-score, tumor size, US-reported LN status, and ABUS retraction phenomenon. It achieved better predictive efficacy than the nomogram without the rad-score and exhibited favorable discrimination, calibration and clinical utility in both cohorts. Conclusion: We developed an ABUS-based radiomics nomogram for the preoperative prediction of ALN burden in BC patients. It would be utilized for the identification of patients with low ALN burden if further validated, which contributed to appropriate axillary treatment and might avoid unnecessary ALND.

RESUMO

A series of thermo- and light-responsive copolymers of poly (N-isopropylacrylamide) (PNIPAM) and 6-[4-(4-methoxy phenyl azo)-phenoxyl-hexyl methacrylate) (AzoMA) (PNIPAM-b-PAzoMA) were synthesized via reversible addition-fragmentation chain transfer (RAFT) radical polymerization. The resulting copolymers had a narrow molecular weight distribution range of 1.06-1.24, in which M n changed regularly with the monomer concentration. Subsequently, the diblock copolymers were successfully modified on the surface of iron oxide nanoparticles through the interaction between the chemical bonds to prepare Fe3O4@(PNIPAM-b-PAzoMA) nanoparticles. The size of fabricated nanoparticles with excellent thermo-sensitivity and photo-sensitivity was controlled at about 40-50 nm. Cell viability assays suggested that the nanoparticles showed no significant cytotoxicity and potential drug delivery in the tumor microenvironment.

RESUMO

This contribution reports the syntheses, structural analyses and properties of europium (Eu3+)- and terbium (Tb3+)-based coordination complexes of poly(N-isopropyl,N-methylacrylamide-stat-N,N-dimethylacrylamide) (poly(iPMAm-stat-DMAm)) copolymer, named as poly-Eu(III) and poly-Tb(III), respectively. In greater detail, poly(iPMAm85-stat-DMAm15) is first prepared by random copolymerization of N-isopropyl,N-methylacrylamide (iPMAm) and N,N-dimethylacrylamide (DMAm) via group transfer polymerization (GTP). Next, poly(iPMAm85-stat-DMAm15) is used as the polymer matrix for chelating with Eu3+ and Tb3+ cations at its side amide groups, to produce poly-Eu(III) and poly-Tb(III). Their structural characterizations by FT-IR spectroscopy and XPS confirm the formation of polymeric complexes. The study on their fluorescence emission characteristics and luminescence lifetime demonstrates that Poly-Eu(III) shows four strong emission peaks at 578, 593, 622, and 651 nm, which are responsible for the electron transitions from the excited 5D0 state to the multiplet 7FJ (J = 0, 1, 2, 3) states, respectively, and poly-Tb(III) also displays four emission peaks at 489, 545, 588, and 654 nm, mainly due to the electron transitions of 5D4 → 7Fi (i = 6, 5, 4, 3). The luminescence lifetimes of poly-Eu(III) (τpoly-Eu(III)) and poly-Tb(III) (τpoly-Tb(III)) are determined to be 4.57 and 7.50 ms, respectively. In addition, in aqueous solutions, poly-Eu(III) and poly-Tb(III) are found to exhibit thermoresponsivity, with their cloud temperatures (Tcs) locating around 36.4 and 36.8 °C, respectively. Finally, the cytotoxicity study on the human colon carcinoma cells LoVo and DLD1 suggests that the luminescent Eu3+ and Tb3+ in the chelated state with poly(iPMAm-stat-DMAm) show much better biocompatibility and lower toxicity than their inorganic salts.

RESUMO

Immunoassays based on enzyme-labeled antibodies have been widely used in the food safety field. However, the production process of enzyme-labeled antibodies is complicated and the low storage stability limits their application. Herein, antibody-horseradish peroxidase (HRP) co-assembled nanocomposites (AHC NCs) with outstanding advantages such as enhanced stability, lower cost, and substrate affinity were successfully prepared via a one-pot green method. Then the AHC NCs were employed as an alternative to traditional enzyme-labeled antibodies to develop a chemiluminescence enzyme immunoassay (CLEIA) toward Escherichia coli (E. coli) O157:H7. Under optimal conditions, E. coli O157:H7 can be detected in a linear range from 1 × 103 CFU mL-1 to 5 × 106 CFU mL-1, while the limit of detection (LOD) is as low as 2.2 × 102 CFU mL-1 (3σ). A series of repeatability studies showed reproducible results with a coefficient of variation of less than 7%. In addition, the proposed CLEIA was successfully applied to the analysis of spiked samples (tap water) and gave quantitative recoveries from 93.72% to 100.72%. This work demonstrates that the developed CLEIA can be applied as a universal platform for specific detection of diversified analytes.

RESUMO

Complete recognition of necrotic areas during small bowel tissue resection remains challenging due to the lack of optimal intraoperative aid identification techniques. This research utilizes hyperspectral imaging techniques to automatically distinguish normal and necrotic areas of small intestinal tissue. Sample data were obtained from the animal model of small intestinal tissue of eight Japanese large-eared white rabbits developed by experienced physicians. A spectral library of normal and necrotic regions of small intestinal tissue was created and processed using six different supervised classification algorithms. The results show that hyperspectral imaging combined with supervised classification algorithms can be a suitable technique to automatically distinguish between normal and necrotic areas of small intestinal tissue. This new technique could aid physicians in objectively identify normal and necrotic areas of small intestinal tissue.

RESUMO

A psychophysical experiment using 3D printed samples was conducted to investigate the change of perceived color differences caused by two different illuminations and two 3D sample shapes. 150 pairs of 3D printed samples around five CIE color centers [Color Res. Appl. 20, 399-403, 1995], consisting of 75 pairs of spherical samples and 75 pairs of flat samples, with a wide range of color differences covering from small to large magnitude, were printed by an Mcor Iris paper-based 3D color printer. Each pair was assessed twice by a panel of 10 observers using a gray-scale psychophysical method in a spectral tunable LED viewing cabinet with two types of light sources: diffuse lighting with and without an additional overhead spotlight. The experimental results confirmed that the lighting conditions had more effect on the perceived color difference between complex 3D shapes than between 2D objects. The results for 3D and 2D objects were more similar under only diffuse lighting. Current 3D results had good correlations with previous ones [Color Res. Appl. 24, 356-368, 1999; J. Opt. Soc. Am. A 36, 789-799, 2019] using 2D samples with large color differences, meaning that color-difference magnitude had more effect on perceived color differences than sample shape and lighting. Considering ten modern color-difference formulas, the best predictions of the current experimental data were found for CAM02-LCD formula [Color Res. Appl. 31, 320-330, 2006]. For current results, it was also found that predictions of current color-difference formulas were below average inter-observer variability, and remarkable improvements were found by adding power corrections [Opt. Express 23, 597-610, 2015].

RESUMO

An experimental dataset, WCG, was assembled. The set includes 416 pairs of samples that surround 28 colour centres and covers a wide colour gamut. The data were used to test the performance of seven colour-difference models, CIELAB, CIEDE2000, CAM16-UCS, DIN99d, OSAGP, and ICTCP, Jzazbz. Colour discrimination ellipses were also fitted to compare the uniformity of the colour spaces. Different versions of the models were derived to improve the fit to the data, including parametric factors, kL, kC, and a power factor. It was found that the kL optimised CAM16-UCS, DIN99d, OSAGP models significantly outperformed the other colour models. In addition, the magnitude of the colour difference had an impact on visual assessment.

RESUMO

BACKGROUND: Helicobacter pylori is a chronic pathogenic bacteria that causes gastric mucosal damage through various host-related and pathogen-related factors. Thus, a single gene research cannot fully explain its pathogenicity. PURPOSE OF STUDY: It is necessary to establish a Helicobacter pylori pathogenic gene transcription factor regulatory network (TFRN) and study its central nodes. RESULTS: The expression data of Helicobacter pylori pathogenic genes were obtained through GEO Datasets of NCBI. The genes were screened using linear model-empirical Bayesian statistics in R language Limma package combined with the conventional t-test; the results identified 1231 differentially expressed genes. The functional analysis (gene ontology-analysis) and signal pathway analysis (pathway-analysis) of differentially expressed genes were performed using the DAVID and KEGG databases, respectively. The pathogenic gene regulatory network was constructed by integrating transcriptional regulatory element database (TRED); the disease-related analysis of the pathogenic genes was conducted using the DAVID annotation tool. Five pathogenic genes (Nos2, Il5, Colla1, Tnf, and Nfkb1) and their transcription factors (Jun, Cebpa, Egrl, Ppara, and Il6) were found to suppress the host immune function and enhance the pathogenicity of Helicobacter pylori by regulating the host immune system. CONCLUSIONS: This effect was largely mediated via three signaling pathways: Tnf pathway, PI3K Akt pathway, and JakSTAT pathway. The pathogenicity of Helicobacter pylori is closely related to the body's immune and inflammatory system. A better understanding of the correlation of the pathogenic factors with the host immune and inflammatory factors may help to determine the precise pathogenic mechanism of H. pylori infection.

RESUMO

Cancer stem cells (CSCs), also known as tumor-initiating cells, are a sub-population of tumor cells found in many human cancers that are endowed with self-renewal and pluripotency. CSCs may be more resistant to conventional anticancer therapies than average cancer cells, as they can easily escape the cytotoxic effects of standard chemotherapy, thereby resulting in tumor relapse. Despite significant progress in related research, effective elimination of CSCs remains an unmet clinical need. CSCs are localized in a specialized microenvironment termed the niche, which plays a pivotal role in cancer multidrug resistance. The niche components of CSCs, such as the extracellular matrix, also physically shelter CSCs from therapeutic agents. Colorectal cancer is the most common malignancy worldwide and presents a relatively transparent process of cancer initiation and development, making it an ideal model for CSC niche research. Here, we review recent advances in the field of CSCs using colorectal cancer as an example to illustrate the potential therapeutic value of targeting the CSC niche. These findings not only provide a novel theoretical basis for in-depth discussions on tumor occurrence, development, and prognosis evaluation, but also offer new strategies for the targeted treatment of cancer.

RESUMO

Thermo-, pH- and glucose-responsive polymeric nanoparticles are of great interest in developing a self-regulated drug delivery system. The novel core-shell nanoparticles were synthesized by self-assembly of a phenylboronic acid-based block copolymer poly-(N-isopropylacrylamide)-block-poly(3-acrylamidophenylboronic acid) (PNIPAM136-b-PAPBA16) and a fluorescent complex glucosamine-poly(N-isopropylacrylamide)/Eu(III) (GA-PNIPAM)/Eu(III) based on the cross-linking between PBA- and GA-containing blocks in this work. The nanoparticles can be tuned via thermo-induced collapse or glucose-induced swelling at appropriate pH and temperatures; they had an average kinetic radius was about 80nm, and which showed excellent fluorescence. MTT assays revealed the nanocarriers had no significant cytotoxic response of the micelle when it was observed in the cell line over the concentration range from 0.1 to 1000 µg/ml at any exposure times.

RESUMO

A dimeric Keggin-type phosphotungstate (ODA)10[(PW11FeO39)2O]·9H2O (abbreviated as ODA10[(PW11Fe)2], ODA = octadecyltrimethylammonium bromide) was synthesized and investigated comprehensively its antitumor activity on MCF-7 and A549 cells. The dimeric structure and amorphous morphology were characterized by FT-IR, UV-vis-DRS, SEM and XRD. The in vitro MTT assay of ODA10[(PW11Fe)2] showed anticancer activity on MCF-7 and A549 cells in a dose- and time-dependent manner, and the IC50 values for MCF-7 and A549 cells at 48 h were 5.83 µg/ml and 3.23 µg/ml, respectively. The images of the ODA10[(PW11Fe)2]-treated cells observed by inverted biological microscope exhibited the characteristic morphology of apoptosis. Flow cytometric analysis showed cell apoptosis and cycle arrested at S phase induced by ODA10[(PW11Fe)2]. The above results illuminated the main mechanism of the antitumor action of ODA10[(PW11Fe)2] on MCF-7 and A549 cells, indicating that this dimeric phosphotungstate is a promising anticancer drug.

RESUMO

All color-difference formulas are developed to evaluate color differences for pairs of stimuli with hairline separation. In printing applications, however, color differences are frequently judged between a pair of samples with no separation (NS) because they are printed adjacently on the same piece of paper. A new formula, ΔENS, has been developed for pairs of stimuli with NS. An experiment was conducted to investigate the effect of different color-difference magnitudes using sample pairs with NS. 1012 printed pairs with NS were prepared around 11 CIE recommended color centers. The pairs, representing four color-difference magnitudes of 1, 2, 4, and 8 CIELAB units were visually evaluated by a panel of 19 observers using the gray-scale method. Comparison of the present data based on pairs with NS, and previously generated data using pairs with hairline separation, showed a clear separation effect. A new color-difference equation for the NS viewing condition (ΔENS) is proposed by modifying the CIEDE2000 formula. The separation effect can be well described by the new formula. For a sample pair with NS, when the CIEDE2000 color difference is less than 9.1, a larger color difference leads to a larger lightness difference, but has no effect on the chromatic difference. When the CIEDE2000 color difference is greater than 9.1, the effect is the opposite. The new formula is recommended for future research to evaluate its performance in appropriate applications.

RESUMO

A strategy for preparing composite micelles (CM) containing both cisplatin(IV) [CisPt(IV)] prodrug and capecitabine using a co-assembly method is described in this study. The CM are capable of an effective release of the anticancer drug cisplatin(II) [CisPt(II)] and capecitabine via acid hydrolysis once they are internalized by cancer cells. Moreover, the CM display a synergistic effect in vitro and the combination therapy in the micellar dosage form leads to reduced systemic toxicity and enhanced antitumor efficacy in vivo.
